Abstract

Rationale. It is known from the literature that the severity of circadian dysregulation is associated with greater severity of psychopathological symptoms and a decrease in the quality of life of patients with schizophrenia. Evaluation of associations of polymorphic variants of the circadian system genes with the severity of schizophrenia symptoms will allow us to approach biologically based stratification of patients and personalize therapeutic approaches. Objective. To identify associations of polymorphic variants of the MTNR1A (rs34532313), MTNR1B (rs10830963), CLOCK (rs801260), PER2 (rs934945) genes with the severity of psychopathological symptoms of schizophrenia. Methodology and Research Methods. The final sample included 390 patients with a verified diagnosis according to the ICD-10 criteria: paranoid schizophrenia (F20.0). The PANSS scale was used to quantitatively assess the severity of psychopathological symptoms in patients with schizophrenia. Genotyping of polymorphisms was carried out using the polymerase chain reaction method in real time. Comparison of quantitative indicators between independent samples was carried out using the Kruskal-Wallis test, subsequently, a regression model was constructed during the analysis. Results. Carriage of the TC genotype of the polymorphic variant of the CLOCK gene rs1801260 was associated with a lower score on item P3 Hallucinatory behavior of the PANSS scale. Conclusion. The heterozygous allelic variant TC rs1801260 CLOCK can be considered a protective factor in relation to the severity of hallucinatory behavior in people with schizophrenia. Given the small effect size, the result should be interpreted as hypothesis-forming, requiring verification and in-depth study in future studies.
